The Detection of Epstein Barr Virus and Human Papilloma Virus DNA in Oral Cancers Using Polymerase Chain Reation


Abstract
The purpose of this study was to observe the Epstein Barr virus DNA, human papilloma virus DNA expression in oral cancer. For this study 91 subjects(45 males, 46 females) are diagnosed as squamous cell carcinoma, salivary gland tumors, odontogenic tumors, spindle cell sarcoma, osteogenic sarcoma and small cell carcinoma referred to Dept. of Oral Pathology. Infirmary, Chosun University were used.
For EBV DNA(EBNA, BNRF-1), and HPV DNA(type 6, 11, 16, 18) detection from the cells, DNA was extracted from paraffin embedded specimens. By the PCR method, EBV DNA, HPV DNA genome were amplified through 30 cycles of denaturation, annealing and extension of primers.
After completion of the PCR, the solution was analyzed by 2% agarose gel electirophoresis. The gel was stained with ethidium bromide and examined by a ultraviolet illuminator.
Obtained results are as follow
1.HPV DNA was not identified in all the subjects.
2.In case of EBV DNA, EBNA gene showed negative reactivity in all the subjects.
3.In case of EBV DNA, BNRF, showed positive reativity in 4 cases(50%) out of 8 cases of salivary gland neoplasms.
4.In case of EBV DNA, EBNA gene, BNRF-1 gene was not appeared on odontogenic tumors(AOT, Ameloblastoma).
The results suggested that HPV DNA did not effect to oral cancer.
